Fixed Field Slide

يعنى ممنوع أن حد يحرك السليد فى الامتحان مسموح بس بلعب فى الفين علشان اختلاف النظر

أولا: السليدز

:emotion-55:

3 questions you ask yourself about the slide

1-Shape (spherical >> Cocci - rod >> Bacilli) spiral just 1 slide will be discussed later isA

2-Color (violet >> Gram +ve - pink "red" >> Gram -ve)

3-Arrangement (pairs = diplococci - chains - clusters )

ONLY the comment we will write in the exam no identification required

as below

slide 1

Posted Image

* ask 1, 2 , 3

spherical , violet gram +ve , clusters

N.B. no pus cells (pink cells) >> in culture

i.e. Comment : "Staphylococcus in culture Gram stain "

* we should mention the stain because we have 3 different stains (gram stain, methylene blue,Ziehl Neelsen )

Test 1

Powerpoint only
Comment:"Single Radial Immunodiffiusion"

Test 2

3 points must write
Comment:"Elek's test ,Negative test ,Non toxigenic C.diphtheria strain"

Posted Image

X crossing shape

3 points must write
Comment:"Elek's test ,Positive test ,Toxigenic C.diphtheria strain"

Test 3

N.B: choose the Highest titre 1/160 as show in 2nd photo
Comment :"Brucella agglutination test showing Zone phenomenon with antibody titre ?? "

Posted Image

Test 4

Widal Test
Very v.v.v.v. Important must STUDY & must SEE in the exam

This test consists of 4 rows of test tubes (3 rows in the same holder "H antigen" + 1 separate row in another holder "O antigen" OR 4 rows in the same holder with 5 openings empty row to separate between O & H antigen )

By Practice >>> will be easier & see this tests
:thnk0001:

Posted Image

O antigen clear , 3 rows H antigen all turbid
Comment:"Widal test: post vaccination"
N.B no titre

Posted Image
O antigen turbid ,get highest titre from it ,
see which row turbid of 3 H antigen name of organism of infection at the control tube
Comment:"Widal test :recent infection with S.typhi "for example" with antibody titre ?? "

Posted Image

If O antigen clear,
see which row turbid of 3 H antigen name at the control tube ,,get highest titre from it
Comment:"Widal test :old infection with S.typhi "for example" with antibody titre ?? "

:emotion-30: :emotion-30: :emotion-30: :emotion-30: :emotion-30: :emotion-30:

Some notes may help
Steps :
1-write Widal test in your exam paper
2-see "O antigen" +ve > turbid tubes > Recent (infection or vaccination "not in our exams")
-ve > clear tubes >> Old (infection or post vaccination )
*** Old > O -ve , H +ve 1 row turbid >> see H antigen turbid to get titre "infection"
*** Old > O -ve , H +ve 3 rows turbid >> NO titre required to mention "vaccination"

3-see last turbid tube highest titre = ?? in O antigen row (in case of +ve O antigen)

4-Organism caused infection (salmonella typhi, paratyphi A, paratyphi B) > see 1st tube turbid or not in the 3 rows
see last turbid tube highest titre = ?? BUT in H antigen rows
To know organism see the "CONTROL" tube = last separate tube in the 3 row holder H antigen
